A Method to Optimize the Excitation Light Sheet in Real Time in Selective Plane Illumination Microscopy

Method to select and adjust the exciation light sheet dimension in a selective plane illumination microscope/light sheet in real time. Background: Selective plane illumination microscopy (SPIM) allows rapid 3D live fluorescence imaging on biological specimens with high 3D spatial resolution. In current SPIM system, the excitation light sheet is selected and optimized before the imaging experiment and it remains the same through the entire experiment, and it is very difficult to change. Technology Overview: Maximizing the benefit of SPIM is due to the excitation light sheet selected and its dimension optimized based on the specimen to be imaged and the desired imaging performance. The excitation light sheet can be tiled quickly within the detection image plane so that the imaging FOV can be extended without sacrificing the axial resolution and the optical sectioning capability of the microscope. Different excitation light sheet are selected to match a given dimension and changed in real time to allow quick optimization of the imaging performance. The excitation light sheet and its dimension can be changed by changing the intensity profile. Advantages: Technique will improve imaging performance of current selective plane illumination microscopy technique. Provides a quick and easy way to optimize the microscope at real time. Applications: Technique increases the flexibility of the selective plane illumination microscopy significatly.
